SRX21221950: Metagenome of cold geyser Wallender Born
1 ILLUMINA (NextSeq 500) run: 105M spots, 31.5G bases, 12.2Gb downloads

Design: DNA extraction from filter samples via Powersoil Max kit of qiagen, library preparation via Westburg NGS DNA Library Prep Kit (cat. no. WB 9096) without PCR amplification. Insert size validation to be more than 400 bp. Sequencing with 2x 150bp paired-end reads on a NovaSwq500 platform to an approximate sequencing depth of 20 Gbp.

Study: Genome-resolved metagenomics of the cold-water geyser Wallender Born

In this project, a 12-day time series of the cold-water geyser Wallender Born in the Volcanic Eiffel (Germany) was sampled. The recovered water was filtered sequentially on 0.2um and 0.1um pore size filters. Additionally, a bulk 0.1um pore size filter sample was taken to recover the entire community in a single fraction. The DNA extracted from the filter samples was to illumina sequencing, followed by genome-resolved metagenomics.
Sample: groundwater was collected from the erupting geyser and filtered sequentially on 0.2um and then 0.1 um pore size filters. Geochemical parameters were determined in duplicate and values depicted here are averages. 0.1µm and 0.2µm samples have same biochemical parameters. Some samples were taken as bulk filters, i.e., direct filtering on 0.1µm pore size filters without previous 0.2 µm pore size filtering to capture the entire community.
